Abstract Background: Odontogenic tumors are a variety of benign and malignant lesions developed exclusively on the jaws. They are known to have variable clinical behaviors and different histological features. The development of odontogenic tumors possibly results from genetic defect or some changes in biological molecules. Aim: To assess the Glypican-3 immune expression in tumor cells of Ameloblastoma, Ameloblastic Fibroma, and Ameloblastic Carcinoma, and correlate its relation with the known biological behavior of these tumors. Also, we were studying nature and arrangement of collagen fibers surrounding tumor cells using picrosirius red stain under polarized light microscope. Methodology: 40 cases were selected from oral and maxillofacial pathology department (ten cases in each group). Sections are prepared to be stained with H&E stain, Glypican-3 immune stain and be examined under light microscope. Other sections are prepared to be stained with picrosirius red stain and to be examined under polarized light microscope. Histomorphometric analysis was done and measure area percents of Glypican-3 immune expression. Also, area percent of red, yellow, and green color separately were calculated from picrosirius red stained sections by image analyzer. Results: level of expression of Glypican-3 is highly significant difference between the groups. The level was highest in Ameloblastic carcinoma group followed by Ameloblastoma group, then Ameloblastic fibroma and lowest in Dental follicle group. Regarding picrosirius red stain results the ANOVA test revealed that the difference between the 4 groups in red, yellow, and green color was highly statistically significant. Conclusions: Glypican-3 immune expression is statistically significant difference among study groups. which may play a role in their pathogenesis and is correlated with their biological behaviors. Different color birefringence of collagen fibers surrounding odontogenic tumors revealed the impact of types of collagens surrounding tumor on its biological behavior.
