Abstract Assessing the protective effect of chitosan and mannan against aflatoxins (AFs) in broilers Coop 500. In addition, change in biomarkers, antioxidant and genetic parameters is the basal objective of this study.For this purpose; aflatoxins from an Aspergillus flavus strain was prepared and the AFs identification and quantification were done by using HPLC analysis.one hundred one-day-old healthy broiler chickens were divided into five groups of ten birds each for 10 and 20(group I –(Negative control group) (fed balanced compound feed according to the age)-group II – (+ve control group) the feed of birds was supplemented with aflatoxin (100 ppb kg-1 diet )-group III – (AF+mannan group) – the feed of birds was supplemented with aflatoxin (100 ppb kg-1 diet) + mannan(1 g kg-1 diet)-group IV – (AF+ chitosan group)– the feed of birds was supplemented with aflatoxin (100 ppb kg-1 diet) + chitosan (0.5 g kg-1 diet)-group V – (AF+ mannan+ chitosan) the feed of birds was supplemented with aflatoxin (100 ppb kg-1 diet) + mannan(1 g kg-1 diet)+ chitosan (0.5 g kg-1 diet). At the end of the experimental period the following analysis was done Kidney function, Liver function, Cholesterol, Free radicals and lipid peroxidation in hepatic tissue, Gene expression for caspase-3 and IL-1 β by RT-PCR, Histopathological examination and HPLC analysis for AFs in liver, muscles and kidney . The results shows that after 10 &20 days poultry that received mannan or chitosan or mannan and chitosan has enhancemaent in growth performance, heamatological parameters ,liver functions ,kidney functions and antioxidant enzyme activity compared to positive control group, also there is reducing in the expression of pro-inflammatory cytokine genes(IL-1β) and apoptotic dysregulation Caspase-3.,also the hepatic parenchymal architecture of the treatment groups (chitosan, mannan, and Chit/MOS) showed a considerable improvement, along with mild vascular congestion. |
