Abstract Immunoassays are divided into two basic groups: I those that used a radioactive label ( isotopic assays) and ii those that use a nonradioactive label (nonisotopic assays). They can also be divided on the basis of whether the tracer or label is on the antigen (RIA) or on the antibody (IRMA). The RIA technique was generally used for the determination of the concentration of virtually substance in small volumes of biological fluids directly without prior pretreatment e.g solvent extraction or liquid solid separation. Thus, many samples can be rapidly tested in large numbers with great sensitivity and accuracy.Measurement of Thyroglobuline (Tg) in serum is an important tool for the diagnosis of thyroid dysfunction, and for monitoring the efficacy of thyroid hormone replacement therapy. Double antibody (DA) radioimmunoassays ( RIA) have considerable appeal because of their extreme sensitivity, greater simplicity and reproducibility. The lack of the literature with respect to the details of the RIA techniques, and the increased consumption of thyroid hormone kits in Egypt, necessitate local development, establishment of an accurate assays and production for the liquid phase (DA) RIA technique for Tg hormon. Hence, the factors affecting all the steps of the RIA technique for suitable, accurate and convenient assays of Tg hormone are critically carried out. This will effectively minimize the cost per test for patients and allow overcoming the cost and logistics of importing the kits of Tg hormone. |
