Abstract That essay is a trial to identify the role of oncostatin M as a therapeutic and diagnostic factor, oncostatin M, is a 28 Kd glycoprotein purified from the conditioned media of phorbol 12-myristate 13-acetate treated U937 cells, based on its ability to inhibit growth of certain tumor cell lines (A375 human melanoma cells) but not normal fibroblasts. Oncostatin M is a distinct member of a cytokine family whose members include interleukin-6, leukemia inhibitory factor, granulocyte colony stimulating factor, ciliary ne~otrophic factor, myelomonocytic growth factor, and cardiotrophin-l. Oncostatin M shows a wide variety of biologic functions on various tissues and cells including, growth modulation, leukemia cell differentiation, low density lipoprotein receptor upregulation, stimulation of plasminogen activator, induction of acute phase proteins, mitogenic activity of AIDS-Kaposi’ sarcoma, differentiation and maturation of megakaryocytes and preservation of RBCs count in irradiated mice, regulation of an extrathymic T-cell development pathway, proinflammatory mediator, regulation of early gene expression and induction of vasoactive intestinal peptides specific receptors. The observation of the mechanisms by which oncostatin M can induce its biological effects and most prop ably through its signal transducer glycoprotein 130 which is shared by other interleukin-6 family cytokines. Glycoprotein 130 is an essential fraction for LIF/OM receptor, and also for specific oncostatin M receptors. |
