Abstract Combinations of NOS inhibitors with antiinflammatory drugs may result in potentiation of their antiinflammatory /antinociceptive effects. This may be due to the additional inhibitory effect, offered by NOS inhibitors, on prostaglandins synthesis and lipid peroxidation process. Inhibition of the cardioprotective effects of endogenous PGs and NO may probably account for the cardiac toxicity caused by a combination of ASA (500 mg/kg) with LNAME (300 mg/kg) in rats. Combinations of low doses of Larg with antiinflammatory drugs may potentiate their antiinflammatory /antinociceptive effects. In the scope of our data, the beneficial effect produced by these combinations in this study can<U+2019>t be attributed to the reported antioxidant effect of nitric oxide. Further studies, perhaps on a molecular level, should be conducted to elucidate the mechanism(s) of action of these combinations. Combinations of higher doses of Larg with antiinflammatory drugs may ameliorate their antiinflammatory /antinociceptive effects. This may be explained by increased PGs synthesis and lipid peroxidation (during inflammation) or activation of NOcGMP pathway (during hyperalgesia). |
