Abstract

The virus disease, in particularly, faba bean necrotic yellow virus (FBNYV), bean yellow mosaic virus (BYMV); banana bunchy top virus (BBTV) and zucchini yellow mosaic virus (ZYMV) were considered as ones of the most important viruses which cause considerable losses in yield productivity as well as the quality of these crops, respectively. The present investigation was conducted to detect viruses infecting faba bean, banana and squash following by identification of them at the biological, serological and molecular levels for designing high sensitive and specific diagnostic tools. Samples of FBNYV; BYMV; BBTV and ZYMV­infected plants judged by ELISA were photographed in the field. The spread of FBNYV and BYMV varied from location to another as 41.8; 58.5 and 51.47% were found in Qalubia, Dakahlia and Dammitta, respectively. DAS­ELISA was used as a diagnostic tool for detecting the presence of BYMV and ZYMV while I­ELISA used for FBNYV and BBTV in the collected samples using polyclonal and monoclonal antisera, respectively. The experimental results confirmed the presence of tested viruses in the infected samples as obtaining ELISA values were over than two folds of the healthy samples. The presence of amorphous inclusion bodies induced by BYMV was tested by the light microscope using 0.5% MGP­Y. The ultrathin sections of BYMV­infected faba bean plants were examined under the electron microscope and cylindrical inclusions were observed as pinwheels and laminated aggregates, in addition phytoplasma like organisms (PLO) with large numbers and different shapes were found. While, the inclusion bodies associated with ZYMV appeared as pinwheel, bundles and scrolls. In addition, the disorganization of plastids, mitochondria and nucleus in cells infected with FBNYV was observed. Filamentous virus­like particles (VLPs) measuring 770X13 nm and 750X13 nm were successfully purified from BYMV­infected faba bean and ZYMV­infected squash plants, respectively, based on the use of polyethylene glycol (PEG­8000).
