Abstract

The protective effects of glutathione (GSH) and granulocyte colony stimulating factor (G­CSF) on myelosuppression induced by 5­fluorouracil (5­FU) were compared in female mice. Animals were allocated to seven groups , the first three were 10/group while the last four were 20/group and were divided into 2 subgroups each. Groups 4a and 4b received 5­ FU in a single dose of 160mg/kg by intraperitoneal route Groups 5a and 5b received 5­FU and GSH in a daily dose of 800mg/kg for 1 week by intraperitoneal route. Groups 6a and 6b received 5­FU and G­CSF in a daily dose of 250 ug/kg subcutaneously for 1 week.Groups 7a and 7b received 5­FU, GSH and G­CSF. The (a) groups were killed after 1 day of 5­FU while (b) groups were killed after 1 week of 5­FU. In groups 5 and 6 protection was demonstrated regarding neutropenia, bone marrow cytology and histopathology, while groups 7a and 7b revealed no protection at all. Therefore, GSH and G­CSF have preventive effects against 5­FU induced haemopoietic toxicity, while combination between them showed no protection.
