Abstract

It is assumed that drugs administered topically to the eye are rapidly absorbed and available at a desirable site in the globe of the eye to exert their therapeutic effect. The main objectives in this investigation were; to prepare and evaluate some ocular formulations that may possibly used in treatment of certain eye diseases. In this thesis, two drugs were selected; the first drug was ketotifen fumarate (An antiallergic agent) and the second one was ketorolac tromethamine (An anti­inflammatory agent). The work in this thesis includes the preparation of some new ocular formulations including ophthalmic DROPs, gels, ointments and ocuserts using different drug carriers. Ketotifen fumarate was prepared in different eye DROPs, gels using sodium carboxymethylcellulose and polyvinyl pyrrolidone; grade k30, and ophthalmic ointments using fatty base, absorption base, Polyethylene glycol base, and Polyvinyl alcohol base. In addition, to investigate the release characteristics, stability of ketotifen fumarate from ophthalmic DROPs, gels and ointments. Further more, the effect of the drug on the non­infectious red eye conjunctivitis was studied. Also the work in this thesis includes, the formulation of ketorolac tromethamine in different ophthalmic preparations; DROPs, gel, ointments and ocuserts. Then, investigate the release characteristics and stability of ketorolac tromethamine from ophthalmic preparations. Also, ocular bioavailability of the drug from different ophthalmic preparations in rabbits eyes tissues and aqueous humor was studied. The obtained results revealed that, ketotifen fumarate can be formulated in these bases. Also, drug amounts released from sodium CMC eye DROPs and gels were higher than that released from PVP K30 eye DROPs and gels respectively. Also, the type of the ointment base could affect greatly the amount of the drug released in the following order, PVA ointment > Water soluble ointment >Absorption base ointment > Fatty base ointment. The average decongestion times of inflamed rabbits eyes treated with the selected formulations are markedly influenced by the type of the drug carriers used. They can be arranged in the following order; PVA ointments > sod CMC eye DROPs > sod CMC gel. Also, the obtained results revealed that, ketorolac tromethamine can be formulated in different stable ophthalmic preparations; DROPs, gel, ointments and ocuserts using different bases like; sodium carboxymethylcellulose, polyvinyl alcohol, hydroxypropyl methylcellulose, carbopol 934, and gelatin. from the obtained results, the drug released from eye DROPs was higher than that from ocuserts and followed by gels. The drug showed a great stability up to 6 months of storage at different temperatures. Ketorolac tromethamine was found in conjunctiva in a higher concentration than cornea. While iris­ciliary body and aqueous humor at all time intervals showed lower concentration
