Abstract •Vitamin D deficiency is a major unrecognized health problem. Not only does it cause rickets in children, osteomalacia and osteoporosis in adults, but may have long lasting effects. •Chronic vitamin D deficiency may have serious adverse consequences, including increased risk of tuberculosis, leukemia, cancers of the colon, prostate, breast,, type 1 diabetes, rheumatoid arthritis, inflammatory bowel diseases, multiple sclerosis, disturbed muscle function and hypertension. •The only way to determine whether a person is vitamin D sufficient, deficient, or intoxicated is to measure the circulating concentrations of 25(OH)D. •1,25- (OH)2D3 and its synthetic analogs are being increasingly recognized for their potent antiproliferative, prodifferentiative, and immuno modulatory activities with super agonistic potency compared with their calcemic effects. •Potential therapeutic applications of VDR ligands are indicated in many diseases including in inflammation (rheumatoid arthritis, psoriatic arthritis), dermatological disorders (psoriasis, actinic keratosis, seborrheic dermatitis), osteoporosis (steroid-induced osteoporosis), cancers (myelodysplasia, leukemia, squamous cell carcinoma, prostate, colon, breast), secondary hyperparathyroidism and autoimmune diseases (systemic lupus erythematosus, type I diabetes, multiple sclerosis, and organ transplantation). |
