Abstract

Potato virus Y considers high risk on potato production hence its effect on potato yield up to 90%. While tobacco ring spot virus (TRSV) risk on potato production consider less than PVY because its effect on potato yield up to 10% only. This study has been done on PVY and TRSV, concerned some trials which were done: 1- Identification Procedures. The identification were based on the symptoms developed on indicator plants, physical properties, serological reactions with the specific antiserum using Indirect -Enzyme Linked Immuno Sorbent Assay (I-ELISA), electron microscopy (EM) negative staining technique, cytological examination using electron microscopy of infected Solanum tuberosum CV. Spunta plants.2- Purification and determination of the molecular weight of PVY and TRSV. The yield of purified PVY was about 4.69 mg virus/ 100 g infected tobacco leaf tissues, while purified TRSV yield was about 5.02 mg virus/ 100 g infected tobacco leaf tissues. 3- Production of polyclonal antiserum against PVY. The PVY antiserum titer was 1/1000 when determined by using (I-ELISA). 4- Maintenance and micropropagation of virus free Solanum tuberosum CV. Spunta plantlets from virus free source. M-I medium was the best medium for establishing shooting cultures. This medium produced big number of shoots and also big size of shoots of potato plantlets according to M-2 and M-3 so we use it as abasic medium for microtuberization. 5- Microtuberization of virus free S. tuberosum CV. Spunta plantlets. The microtubers formation begins after one week from transformation. Average number of microtubers per plantlet was 4.9 microtubers after 30 days from transformation.
