Abstract The thesis is devoted to the pharmaceutical applications of different analytical methods for the determination of certain antiviral drugs, namely, acyclovir and ribavirin. The proposed methods were successfully applied for the determination of the studied compounds in their pharmaceutical formulations. Two spectrophotometric methods were developed for the analysis of acyclovir and ribavirin, based on the oxidative coupling reaction of3-Methyl benzothiazolin 2-one hydrazone hydrochloride with the studied drugs after their oxidation using ceric ammonium sulphate or potassium persulphate in acid medium to produce a deep blue color peaking at 630nm. Simple and sensitive kinetic methods for the determination of ribavirin in bulk and in its pharmaceutical preparations using alkaline potassium permanganate as an oxidizing agent were developed. The methods are based upon a kinetic investigation of the oxidation reaction of the drug at room temperature for fixed times of 20 and 30 minutes. In the first method, the absorbance of the colored manganate ion was measured at 610 nm, while in second method the reduction in the absorbance of permanganate was measured at 525 nm. It aims to develop and validate a stability indicating, and rapid method for the determination of the antiviral acyclovir, based on its polarographic complexation with Nickel (II) and measuring the produced pre-wave. The proposed method depends upon studying the polarographic activity of Nickel (II)-acyclovir complex in Britton Robinson buffer (BRb) at pH 5. A simple polarographic method for the determination of ribavirin using complex formation technique. It aims to develop and validate a stability indicating, and rapid method for the determination of the antiviral ribavirin, based on its polarographic complexation with Nickel (II) and measuring the produced catalytic pre-wave. The proposed method depends upon studying the polarographic activity of Nickel (II)-ribavirin complex in 2.5 X 10-3 M HCl. The proposed methods were applied successfully for the determination of the cited drugs in their pharmaceutical formulations. The validation criteria of the developed methods were intensively studied. All the results were statistically analyzed and compared with those given by official methods. |
