Abstract

Bleomycin is well known by its antitumor activity both in vitro and in vivo. However, moderate nausea and vomiting occur in virtually all patients taken BLM. Hyperpyrexia was also reported in some cases. In the present study, the combinations of BLM with ondansetron and/or paracetamol were evaluated in experimental ascites carcinoma model in vitro and in vivo by induction of solid tumor in mice via S.C. injection of EAC cells. Ondansetron and/or paracetamol did not alter the antitumor effect of BLM in vitro and in vivo. However, ondansetron prolonged the survival time in BLM/ondansetron-treated animals. The lung toxicity of BLM was evidenced by decrease in the body weight, increase in the lung/body weight ratio, decrease in the response of pulmonary arterial rings to 5-HT and increase in the contractility of tracheal smooth muscles induced by ACh. The toxicity was also confirmed biochemically by marked increases in hydroxyproline and lipid peroxidation in rat lung and the decrease in GSH level. Prior treatment with ondansetron decreased lipid peroxidation and normalized GSH level and hence enhanced the percent survival of rats. Concomitant use of paracetamol did not alter any of the measured parameters. Administration of both ondansetron and paracetamol with BLM increased the contractility of tracheal smooth muscles significantly as compared to BLM alone, elevated SOD activity to a significant level. The percent survival of animals decreased to 50%. So ondansetron or paracetamol can be used freely with BLM, while use of both drugs with BLM should be avoided.
