Abstract

Owing to the great importance of steroid drugs, alysts spent tremendous efforts for the development highly selective and sensitive methods for their t1fication either in bulk or in various dosageThe objective of the present work is to develop a high selectivity and sensitivity or some chosen steroid drugs: Ethinylsteroids as ethinyloestradiol, mestranol, norethisterone, norethisterone acetate and norges- irel. Corticosteroids as cortisone, cortisone acetate, hydrocortisone, hydrocortisone acetate, prednisone, pred1nsolone, betamethasone, dexamethasone and triamcinolone acetonide. Methyltestosterone. This part is devoted” for developing a colorimet-
ric assay for 17 o<.-ethinylsteroids.
The procedure
is based on the conversion of the drugs to l7-keto derivatives by sodium but oxide in non aqueous medium. Then the de-ethinylated steroids are subjected to Zimmermann reaction with either 1,3-dinitrobenzene or l,3,5-trinitrobenzene. The colour developed is then measured at Xmax 515 nm and 475 nm respectively. The method is sensitive and selective for the steroids containing 170( -ethinyl group. The mean percent recoveries obtained range from 99.63+1.0322lOO.04:±.O.74l.

This part describes the utility of oxalyldihydraazide for the colorimetric determination of certain
corticosteroids via their C-17 side-chain. The drugs quantitatively release an equivalent amount of fonn~ aldehyde upon oxidation with sodium bismuthate. Formaldehyde reacts with oxalyldihydrazide in neutral medium in presence of copper (11) ions producing a highly coloured condensat ion product, which absorbs imax 610 nm. The reaction obeys Beer’s law in -1

This part involves a study of the reaction of A4-J-ketosteroids with certain hydrazine derivatives U 1sonicotinic acid hydrazide, thiosemicarbazide, em1carbazide and oxalyldihydrazide.The reaction prooducts absorb appreciablly in ultraviolet region with a considerable high molar ab sorpt i vi ties.
The conditions required for stable reaction prooducts are thoroughly studied, and a procedure is deveeloped to estimate the drugs either in bulk or in dosage form by each reagent respectively. The results obtained are statistically compared 1th those of the isonicotinic acid hydrazide method, where both methods proved to be equally accurate and precise.
This part includes the official procedures used the determination of studied compounds.
