Abstract

The incidence of TB is increasing worldwide and recent reports of MDR-TB have stimulated the search for new antituberculous agents. Fluoroquinolonesare the most recent class of antibiotics offering a hope in the fight against thisdisease.
This study aimed to determine the susceptibility of M.tuberculosis clinicalisolates to 2 of the fluoroquinolones: CIP and OFX in comparison to the 2 key.components ()idle first line drugs. IN1-1 and Rif.
This study was conducted on 100 cases suspected of having pulmonarytuberculosis based on full history, clinical manifestatiOns and radiologicalexamination. The patients had never received antituberculosis treatment withage ranged from 18-75 years.
Morning sputum samples on 3 successive days were collected from eachpatient and subjected to:-
I-Direct smears stained by Ziel-Neelsen stain.
2-Digestion-decontamination procedure using (NALC-NaOH) method.
3-Cultivation on L .1 media.
4- Identification of isolates by ZN stained films and the biochemicalreactions: niacin accumulation, nitrate reduction and heat stableCatalase tests.
The susceptibility of M.nthcrctdosis isolates to Rif, INII, CIP and OFXwere determined by the conventional proportion method on middle brook 71-1,,,(MB71-11(1) agar.
The 100 sputum samples revealed 38 positive cultures, 20 of them werepositive by direct kinyoun’s stain and 29 by kinyoun’s stain after processing.The sensitivity of direct smear versus LI culture was 52.6% while the sensitivityof stained smear after processing was 76.9%.
