Abstract

The work presented in this thesis involves preparation and evaluation of formulation containing drugs for localized treatment of periodontitis. The need for localized treatment originated due to the side effects which were observed to be accompanying the systemic treatment of periodontitis. The percent drug released of the drug was tried to be enhanced by incorporation o0f penetration enhancers. Also evaluation of the effects of some of the prepared formulations on the healing of periodontitis induced experimentally in guinea pigs (48 animals) which were selected as experimental animal model. The animals are divided into one control group and other are treated group. The animals were killed at different time intervals representing the healing at different stages To fillful the objectives of this studies the thesis was divided into two main parts Part I: Formulation and Evaluation of Clindamycin Hydrochloride Bioadhesive Matrices In this part, clindamycin hydrochloride was formulated in bioadhesive tablets and gels. Penetration enhancers were added to gel formulations and their effects on the physicochemical and release properties of the drug were investigated. The effects of clindamycin hydrochloride selected formulations on the healing of experimentally induced periodontitis were also studied Formulation and Evaluation of Metronidazole bioadhesive matrices Part II: Metronidazole was formulated in bioadhesive gels and films. Menthol and oleic acid were added and their effects on the physicochemical and release properties of the drug were investigated. The effects of metronidazole selected formulations on the healing of experimentally induced periodontitis were also evaluated It was found that clindamycin hydrochloride could be formulated in the form of bioadhesive tablets and gels and on the other hand metronidazole could be formulated in the form of bioadhesive gels and films. Also it was revealed that the release of the drug could be enhanced by incorporation of penetration enhancer. For both clindamycin hydrochloride and metronidazole, it found that the formulations tested on the periodontal tissues of guinea pigs enhanced the healing and so they could be used for localized treatment of periodontal diseases.
In the second part was formulation assessment gels and adhesives vital chips containing to Akaa metronidazole. The study was vital adhesion force and the rate of the drug from the starting formulations stomach was also study the effect of some selected formulations to heal the tissues supporting the teeth after the introduction of the disease in the tissues of guinea pigs.
To achieve the objective of the study included the two parts:

This section deals with the formulation of clindamycin hydrochloride drug in the form of tablets and gels, adhesives vital. Has been added incentives for gel formulations and study their impact on the physico-chemical properties and starting the medication were also study the effect of some selected formulations to heal the tissues supporting the teeth after the introduction of the disease in the tissues of guinea pigs.
Therefore split work in this part into two classes:
Chapter I: formulation and evaluation of tablets and gels, adhesives containing vital drug clindamycin hydrochloride.
This chapter has been divided into:
Chapter I (a) the formulation and evaluation of adhesive disks containing vital drug clindamycin hydrochloride.
Was prepared adhesive disks containing vital CR particles following Krbopoul the 943 hydroxy Brobbea-methyl cellulose and sodium carboxy methyl cellulose and sodium different genes Petrkizin the 50 and 100 mg. Clindamycin were merged to hydrogen chloride at a dose of 50 mg in the middle of Mannitol or Alavasal. The study of the physico-chemical properties of the tablets were also Bunting vital adhesion force and surface pH as well as the swelling properties as well as studying mechanical rate starting medication.
