Abstract Protease enzymes account for a significant proportion of the world enzyme market and are used in a variety of processes that including food and biotechnology industries. Recently, the microbial sources play a great role in enzymes production. Therefore, this study aimed at production of protease en7.yme using microorganisms. The microorganisms producing protease enzyme were isolated from soil to obtain bacterial strains producing high yield of protease. These bacterial strains were identified. The study also included the effect of various environmental and nutritional factors on growth and protease production by these strains. The produced protease were partially purified and characterized. These enzymes were industrially applied to study the effect of this enzyme on the functional properties of protein isolates from both plant and animal origin. The results obtained are summarized as follow:- 5.1. Isolation of microorganisms producing protease enzyme; Microorganisms were isolated from soil samples by enriching thegrowth medium such as casein agar medium. Isolates were identified as bacillus strains. Three of these isolates showed high protease activity, which were identified as Bacillus foams , Bacillus PumiluS and Bacillus lichenformis. 5.2. Production Of protease: 5.2.1. Tested media: Induction media gave the maximum protease activity for three Bacillus strains followed by glucose casein mineral salts medium (CMSM), bran extracted peptone broth (BEPB) and nutrient casein broth (NBC). 5.2.2. Environmental conditions: The optimum induction temperature to produce maximum growth and protease yield by Bacilllf, fumuiS were 45 and 40°C after 48 hrs incubation, respectively, by Badllus pumutslls were 40 and 40°C after 24 hrs incubation and by Badlllls lichenl!olmis were 45 and 45°C after 4& hrs incubation, respectively. |
