Abstract

Male rat were divided to 8 groups. Control group taking saline by intragastric tube for 60 & 90, valproate group taking drug by intragastric tube for 60 & 90, lamotrigine group taking drug by intragastric tube for 60 & 90 and topiramate group taking drug by intragastric tube for 60 & 90. It had been found that lamotrigine exhibited an extreme toxic effect on rat testis as it caused total degeneration of the seminiferous tubules and absence of sperms in their lumen. While, valproate leads only to presence of few vacuoles in seminiferous tubules, and topiramate caused total degeneration of some tubules but other tubules were completely normal. So, caution must be taken when lamotrigine is used in male patients. On the other hand, lamotrigine did not cause congenital malformation in any of the newborns but, valproate caused marked weight reduction and topiramate appeared to have a teratogenic effect as one case of intestinal herniation through abdominal wall. Therefore, it is advisable not to treat pregnant women with neither valproate nor topiramate. However, more studies needed to confirm the safety of lamotrigine on pregnant females.
