Abstract

Chronic inflammatory disease is believed to pose a tremendous medical burden in the developed world, as it causes loss of workers’ productivity. This study investigate the predicted anti-inflammatory effects of simvastatin compared with aspirin, and evaluate the effect of using them in combination which may produce synergistic effect and lower the dose required for each agent. The present study was conducted on 72 male Wistar albino rats, induction of inflammation was done by air pouch method, the animals where divided into 12 groups, they received the anti-inflammaory treatment for either 3 or 6 executive days. Evaluation of lipid peroxidation and antioxidant activity were done by measurement of malondialdehyde and nitric oxide concentrations. Investigation of inflammatory mediators including C-reactive protein, TNF-α, IL-6 and IL-4 were also carried out. There were no significant differences between simvastatin and aspirin treated groups in levels of MDA, NO, CRP, TNF-α, IL-6 and IL-4. Combined therapy possesses better antioxidant activity and better reduction in proinflammatory cytokines. We can conclude that, the anti-inflammatory action of simvastatin is comparable to that of aspirin; also combination of simvastatin and aspirin may produce synergy and have a lot of benefits in immune response. simvastatin can be used instead of aspirin for treatment of inflammation specially in some cases like: Patients with hyperlipidemia, gastric ulcer, bone diseases and neuropathy. Therapies directed specifically against proinflammatory cytokines have to be tested in appropriate prospective clinical trials to investigate their potential in inflammation management.
