Abstract

The rhizome of Ginger (Zingiber officinale Rosc.) has been used as a spice and for medical purposes in Asian, Indian, and Arabic herbal traditions since ancient time. Recently, it has gained considerable attention as a botanical dietary due to its active biological constituents of phenolic substances ([6]-gingerol, and [6]-paradol) that possess anti-inflammatory, and anti-oxidative properties. It also exerts substantial anti-carcinogenic activities. The present study used aqueous extract of Ginger concentration (120mg/ml) as a protective agent against the toxicity of the reproductive system of immature female rat induced by DMBA. This study used DMBA (40 mg/kg body weight) as a potent carcinogen causes destruction of different ovarian follicles to form the ovotoxicity. Histopathological and histochemical observation of DMBA treated group revealed extensive edema of the ovaries, pore ovarian cycle in which all types of the ovarian follicles are degenerated, extensive proliferation deeply in the stroma referred to as stromal hyperplasia may be represented aggregation of transformed and/or cancer cells. The ovarian tissue also exhibit congested blood vessels, large corpora lutea, and disorder distribution of protein, and carbohydrates content in comparing with the control ones. Immunohistochemical observation was patchy in the granulose layers of the most follicles and corpora lutea and showed very high significant increase in PCNA LI in comparing with control and ginger treated group. Ultra-structural observation of DMBA treated group showed the most follicular and fibroblast cells were at different stages of apoptosis. Cytoplasm showed vacuolated and swollen mitochondria, numerous active lysosomes. Primordial follicles exhibited broken nuclei The DMBA and Ginger treated group exhibited a degree of the ovarian tissue improvement in which numerous of developing follicles at different stages are possessed, the protein and carbohydrates content showed moderate amount of distribution in comparing with control group. Immunohistochemical observation of this group showed very high significant decrease of PCNA LI in comparing with DMBA treated group. The ultra-structural of DMBA and Ginger treated group possessed the cytoplasm with electron lucent , increment of the amount of euchromatine material , decreasing of the number of lysosome granules , increasing of the number of lipids droplets, and mitochondria almost at normal structure. Finally, uses of aqueous extraction of ginger may be played a role as a protective agent against ovotoxicity induced by DMBA due to its anti-oxidant and anti-cancer properties.
