Abstract

This study was conducted on a total of 20 trachea and larynx samples collected from clinically diseased birds suspected to be infected with infectious laryngotracheitis virus. Diseased birds were showing respiratory signs and mucoid discharge from the eyes caused the eyelids to become glued together, with drop of egg production in layers. Samples were collected from different farms in Dakahlia and Damietta Governorates. A trial for isolation of ILT virus from trachea and larynx samples via CAMs of 11 days old ECEs was conducted, three to six passages were carried out for each sample depending on the presence of distributed pock lesions. Fourteen samples gave characteristic pock lesions seven samples after 2nd passage, one sample after 3rd passage, 3 after 4th passage and 3 after 5th passage. The isolated virus was identified using agar gel precipitation test, immunoperoxidase, immunoflourescent. Immunoflourescent and immunoperoxidase,techniques were more sensitive than agar gel precipitation test in virus identification. Histopathological examination was also used for detection of syncytial formation and intranuclear inclusion bodies of Gallid herpesvirus-1. PCR could be used as a reliable and specific technique for confirmation of Gallid herpesvirus-1 in infected CAMs.
