Abstract Anther culture technique, as a novel technique for plant breeding, was used in this investigation to understand the ability of embryoid induction and plant regeneration for five different cultivars of the Egyptian wheat (Triticum aestivurn L.). These cultivars were ”Mabrouk”, ”Giza 155”, ”Giza 162”, ”Sakha 69” and ”Sakha 61”. Two different regenerated media were also included in this study, as well as the embryoid induction medium. The main characters that were measured for the tissue culture experiment were: anthers responded for embryoids production, number of embryoids, regenerative capacity, the ability of green plantlets formation and the greenlalbino ratio. The cytological analysis and chron~osomec onstitution study of the androgenetic derived regenerants were carried out. Biochemical analysis of peroxidase isozymes extracted from nonembryogenic, embryogenic callus and young green plantlets were also embodied in this investigation using polyacrylamide gel electrophoresis (PAGE) technique. The obtained results could be summarized as follows: 1. Anther response to tissue culture: The observed results revealed that ”Giza 162” showed a superior trend for the highest value of responded anther, while ”Sakha 61 ” and ”Sakha 69” cultivars showed the lowest trend. 2. Embryoid production: ”Giza 162”, ”Mabrouk”, and ”Giza 155”, cultivars showed the highest values for the number of embryoids, while ” Sakha 61 ” and ” Sakha 69” were having the lowest values. |
