Abstract

The present study has been done to demonstrate the effect of some hypoglycemic drugs (regular insulin, chlorpropamide, metformin and and gliclazide) on the blood glucose, serum c h o l e s t e r o l , serum p r o t e i n s albumin/globulin r a t i o a n d prothrombin time in blood of male albino rats in both normal and in conditions of alloxan induced diabetes by spectrophotometric method. About 300 male ablino rats have been used for this study, these rats were divided into (10 groups) : 1- The first group of 10 normal rats used as a control group of the diseased animals. 2- The second group of normal rats (10 rats) were treated by daily injection of regular insulin. 3- The third group of normal rats (10 rats) were treated by daily oral administration of chlorpropamide. 4- The fourth group of normal rats (10 rats) were treated by daily oral administration of metformin. 5- The fifth group of normal rats (10 rats) were treated by daily oral administration of gliclazide. 6- The sixth group of alloxan diabetic rats (50 rats) without treatment for a period of eight weeks (control group). 7- The seventh group of rats (50 rats) with alloxan induced diabetes and treated by daily injection of regular insulin for eight weeks. 8- The eight group of (50 rats) with alloxan induced diabetes were treated by daily oral administration of chlorpropamide for eight weeks. 9- The ninth group of 50 rats with alloxan induced diabetes were treated by daily oral administration of metformin for eight weeks. 10- The tenth group of 50 rats with alloxan induced diabetes were treated by daily oral administration of gliclazide for eight weeks In the control group of alloxan induced diabetes the blood glucose level was increased all over the period, such high level of blood glucose was maintained by fortifying doses of alloxan one tenth of the initial dose every ten days. On the proper day the animals were sacrificed by decapitation and samples of both plasma and serum were taken from each animal and were submitted for chemical analysis to estimate: 1- Blood glucose by enzymatic determination Trinder’s glucose oxidase method using phenol and 4-aminophenazone (Trinder, 1969) 2-Serum cholesterol by enzymatic determination (Flegg , 1973) 3- Serum proteins, albumin and globulin fractions by the Biuret method (Peters , 1968) and by using Bromcresol Green (Doumas et al., 1971). 4- Prothrombin time, in plasma, by the use of Quick method (1 949).
