Abstract
This study was c a r r i e d o u t a t P l a n t Cell and Tissue Culture Department, Genetic Engineering and Biotechnology ~ivision, National Research Centre, Giza, Egypt, during the period from 1989 t o 1991. This study aimed t o i n v e s t i g a t e t h e following p o i n t s : 1. Establishement o f c a l l i c u l t u r e s from d i f f e r e n t explants of cu’ltlvat~d tomato Lycooersicon esculentun, I.. , cv. Peto 86. The d i f f e r e n t explants i n c l u d i n g hypocotyls, cotyledonary l e a v e s , r o o t s and seeds. Moreover, the procluction of c a l l u s under s a l i n e conditions was induced. Callus produced and maintained on the salire media accuired the name s e l e c t e d c e l l l i n e , while t h a t produced and maintained on s a l t - f r e e media c a l l e d non-selectsd c e l l l i n e . 2. S a l i n i t y s t r e s s studies,including. t h r e e experir~ents: A) The e f f e c t ~f XzC1 saiinity on both selectez and zcn s e l e c t e d c e l l l i n e s . 5) The e f f e c t of Sac1 s a l i n i t y on calli c u l t u r e s Zer-i7:ed frox d i f f e r e n t cx;lants cf tometo seecling. C) The e f f e c t of s a l i n i t y stress r e s u l t i n g from NaCl alone an6 t h a t r e s u l t i n g frorr~ a mixture of salts i n the for2 of a r t i f i c i a l sea water on c a l l u s c u l t u r e s .
