Abstract Monoclonal Antibodies preparation through production of hybridomas secreting specific antibodies against tetanus toxin. The hybridomas has a characteristic feature of both parent cells tha ability to grow indefinitely from myeloma and the ability to produce the specific antibody from the B-cells. In this study the production of monoclonal antibodies against tetanus toxin which have a manyadvantages than the polyclonal antibodies (serum): 1- The monoclonal antibodies have a low tendency to produce hypersensitivity reaction that can caused by the polyclonal antibodies prepared in equine. Monoclonal antibodies production need only for lab to production rather than polyclonal antibodies need farm to produce antitetanic serum in equine. The tetanus toxoid was conjugated with adjuvant to produce a stable emulsion. Primary intraperitoneal immunization of BALB/C mice by using emulsion contain tetanus toxoid conjugated with complete freunds adjuvant, boosters doses used emulsion contain tetanus toxoid conjugated with incomplete freunds adjuvant. The final dose was tetanus toxin only 2-3 days before fusion procedure. |
