Abstract

The present investigation was carried out in the Agronomy Department Faculty of Agriculture, Kafr El-Sheilch, Tanta University, Icafr El-Sheild~. The aim of the present investigation was (1) to define the most suitable medium, the potato-2 medium as a two step culture or the 85D12 medium as one-step culture to evaluate the response of some cultival-s of bread wheat; namely; ”Salfia 8”, ”Salcha 92”, ”Saklia 61” and ”G. 163”; to induce haploid plants through anther culture, (2) to study the response of seven wheat cultivars and one promising line as well as the Fls to in vitro anther culture under three callus induction media (Potato-2, C 17, 85D 12supplemented with 2, 4-D) and each of the two sources of carbon (sucrose or maltose) and one regeneration medium (1 90-2 medium), to identify the best medium and the suitable source of carbon for callus induction and plant regeneration for each genotype. Fourteen wheat genotypes, namely, ”Sakha 8”, ”Saldna 61”, ”Saltlia 69”, ”Sakha 92”, ”Giza 155”, ”Giza 163”, ”Giza 164”, ”Line # 22” and the F1 hybrids ”Giza 163 x Sakha 92”, ”Giza 163 x Saldia 8”, ”Salcha 8 x Salha 61”, ”Giza 155 x Salclia 8”, ”Saltha 69 x Saldia 92” and ”Line #22 x Saldia 92” were used as plant material in this investigation, and Finally, (3). to select salt tolerant dihaploid plants for wheat breeding. The wheat genotypes, ”Sald~a 8”, ”Saltha 92”, ”Giza 163”, ”Line # 22” and the F, hybrids ”Giza 163 x Saldia 8”, ”Giza 163 x Sakha 92” and ”Line # 22 x Salcha 92” were included in the screening procedure. The results obtained throughout this study could by suin~narizeda s follows : Potato-2 as two-step procedures was efficient for ”Giza 163”, ”Saltha 8”, ”Sakha 92” for callus induction and plant regeneration, whereas, 85D12 containing NAA 0.75 ingll and Kinitin 1.5 mgll desinged for one-step procedures culture efficient for ”Saldza 6 1 ”. The analysis of variance indicated that callus induction showed highly significant variations under the influence of the different factors (genotypes, media and sources of carbon) and the interaction between factors. 2.1. ”Giza 164” variety gave the highest nuinber of callus induced calli, whereas the ”Sakha 69 x Saldia 92” cross gave the lowest nuinber of induced callus. 2.2. The potato-2 medium gave the highest number of induced calli (2.36), while, the medium 85D12 gave the lowest number (0.51). 2.3. Sucrose as source of carbon gave the highest mean number of induced calli comparing with maltose that gave the lowest number. 2.4. The effect of genotypes x medium interaction: the analysis of variance indicated that ”Giza 164” gave the highest number of induced calli, when cultured on potato-2 medium, whereas all genotypes gave the lowest number of calli with 85D12 medium, except the cross ”Line /# 22 x Salcha 92” that gave the lowest number of calli with, potato-2 medium (0.76 + 0.39). The effect of medium x source of carbon interaction: the potato-2 medium with sucrose gave the highest number of callus (2.87), whereas the C 17 medium with maltose gave higher induced calli (1.97) compared to that with the other two media. The effect of genotype x source of carbon interaction all genotypes gave the highest response to callus induction with sucrose, except for Sakha 69, Salcha 6 1, Line # 22 gave the highest response to number of callus induction with inaltose. 3- Callus diameter : However, the differences were significant or highly significant, only in soine cases between the three media and the two carbon sources. Potato-2 medium induced bigger callus diameter when maltose was added instead of sucrose with Saltha 92, Giza 163, Giza 155 and Line # 22, while the same medium with sucrose was efficient for Saldia 8, Giza 163 x Saltha 92”, ”Giza 163 x Salcha 8” and ”Line # 22 x Salha 92”. C 17 medium with sucrose was efficient for ”Giza 1 55 x Salcha 8”, while C17 medium with maltose was efficient for Salha 69, Saldia 61 and ”Sald~a 8 x Salcha 6 1” cross. 85D12 medium with ~naltosew as efficient for Giza 164.
