Abstract SUMMARY Schistosomiasis is the most important problem in our country. This work is concerned with the detection of circulating schistosomal antibodies, circulating immmune complexes and deposited schistosomal antigens in tissues of S.mansoni infected mice in a trial to evaluate their potential use in diagnosis of the disease and efficacy of praziquantel therapy. One hundred and thirty mice were infected by tail imersion with 100 (± 10) S.mansoni cercariae for each one and treated by PZQ(500 mg / kg! body weigi] after 8 weeks from infection and investigated every 2 weeks until the 101/1week after treatment. -Thirty five mice were also infected by S. mansoni by the same manner, received no treatment and investigated (nontreated infected group). -Thirty five completely free mice were not infected and investigated (negative control group). The biboratory investigations were done for all mice included in this study: 1- Stool examination by Kato thick smear technique for counting the S mansom eggs. 2- Perfusion technique to count the worm burden. 3- ELISA method for eSAb and eIe detection in the sera. .4-Indirect immunoperoxidase technique for schistosoma! antigen detection in the tissues (liver, spleen, intestine, kidney and heart). 125 I.hUQllowjng finging wcrc obtaincd : I- Highly significant increased levels of CSAb &CIC in the infected mice before and after treatment than the non-infected control(P <O.OOl)were found. 2-Highly significant decreased levels ofCIC after praziquantel therapy were detected in completely cured mice 10 weeks after treatment(P <0.001). 3-Highly significant difference was detected between CSAb & CIC level before and after praziquantel therapy (P<0.02- P<O.OOI). 4-Positive correlation was found between CSAb & CIC before and after treatment (P<O.OI). 5-Highly significant decreased number of eggs and worms among the infected mice after 2 weeks of treatment than before treatment . 6-Positive correlation was found between the egg count and the worm burden before and after treatment (P<O.Ol). 7-Highly significant increased rate of the cure was detected (92.6%) 10 weeks after praziquantel therapy . 8-Highly significant increased level of CSAb was detected after 2 & 4 weeks of treatment than before treatment(P<O.OOI). 9-Insignificant increased level of CSAb was detected after 6 weeks of treatment than before treatment(P>O.05). 10-Insignificant decreased level ofCSAb was detected after 8&10 weeks of treatment than before treatment(P>O.05). II-Highly significant increased level of CIC was detected after 2 & 4 weeks of treatment than before treatment(P<O.OOI). 12-Insignificant increased level ofCIC was detected after 6 & 8 weeks of treatment than before treatment(P>0.05). 126 13-Insignificant decreased level of eIe was detected after 10 weeks of treatment than before treatment(P>O.05). 14-The deposited schistosomal antigen (adult whole worm) could be detected in the livers,spleens and intestines before and after treatment. 15-The amount of deposited schistosomal antigen slightly increased 2&4 weeks after treatment and completely disappeared 10 weeks after treatment. 16-Positive correlation was found between the deposited schistosomal antigen and the egg count before and after treatment(P<O.OI). 17-Positive correlation was found between the deposited schistosomal antigen and the worm burden before and after treatment(P<O.Ol).
