Abstract

The alkylating agents are the largest class of anticancer agents, comprising 12 commonly used drugs and 5 subgroups: mustards, alkylsulfonates, niytoosoureas, ethylemines, and triazenes. In addition, several other agents are thought to act, at least in part, by alkylation, including procarbazine, hexamethylmelamine, dacabozine, estramustine, and mitomycin C. Also alkylating agents are compounds that are cpable of introducing alkyl groups into nucleophilic sites on other molecules through the formation of covalent bonds. These nucleophilic targets for alkylation include the sulfhydryl, amino, phosphate, hydroxyl, carboxyl, and imidazole groups that are present in macromolecules and low- molecular- weight compounds contained within cells.
