Bipolar disorder has a life time prevalence of 1% and is characterized by fluctuations in mood, ranging from extreme elation or mania to severe depression usually accompanied by disturbances in thinking and behaviour. Family, twin, and adoption studies provide strong evidence of important genetic factors but the mode of inheritance is complex and non-mendelian. It results from many genes interacting with each other and with the environment in a multifaceted way, making identification of any gene difficult. Genes known to be involved in biologic mechanisms that can result in disease development are ideal candidates to target for genetic etiological involvement. The serotonergic system is one example of a system for which identification of possible candidate genes and polymorphisms may be important, as this system is known to affect mood, emotion, and cognition. Among genes in this system is SLC6A4 or 5-HTT, which codes for the serotonin transporter. This gene contains two primary poly-morphisms studied by researchers. The first poly-
