Abstract A total of 69 Pseudomonas aeuginosa isolates poultry fish and human were subjected for bacteriological , biochemical characterization , SDS- Page analysis for the isolates showed that Pseudomonas aeuginosa contain a wide variety of proteins ( OMPs) of different M. W. with predominate bands .Plasmids DNA of Pseudomonas aeuginosa isolates were studies some strains have no plasmid while other have one or more plasmids with different M.W. DNA fingerprinting bases on RAPO – PCR technique were studies all the isolates characterized by two common bands. Antibiogram for Pseudomonas aeuginosa isolates towards 9 chemotherapeutic agents showed that all the isolates are sensitive for Ciprofloxacin ( 93%) Norfloxain (90) . Danofloxacine and Olfoxacin ( 88%) while for Gentamycin and Amikacin 80% .On the other hand all the tasted strains were highly resistant to Amoxycillin and Rifampcin (Rimactane ) ( 100%) followed by Amoxyillin + Clavulanic acid ( 93%) . Detection of Norfloxacin gene resistance in Pseudomonas aeuginosa isolates from the different origins the genewas amplified by PCR using two primers in positive samples were showed that the amplified fragment was arounf 960 bp. Exotoxin A ( ETA) gene of Pseudomonas aeuginosa isolates from different orgins were amplified by PCR using two primers , The positive samples for ETA gene showed amplified fragment with M. W 369 bp. |
