Abstract

Three experiments were carried out at The Tissue Culture Lab. of Strawberry and Non-Traditional Crops Center, Faculty of Agriculture Ain-Shams University, during the period from 2003-2005. Virus-free plantlets of three strawberry cultivars, namely: Camarosa, Sweet Charlie and Rosalinda were obtained by tissue culture technique and farther used throughout this study.
The first experiment was conducted to investigate the effect of various concentrations of BA and kinetin as applied to the tissue culture media. Cytokinin induced higher proliferated rates with all used cultivars. The highest number of shoots per explant obtained was 20.7 shoots in Camarosa cultivar when grown on MS medium supplemented with BA at 0.25 mg/L; 345.5% increase compared to control (6.5 shoot/explant). Kinetin induced significantly higher proliferation rate (13.41 shoot/explant) compared to (12.4 shoot/explant) when BA was used (5.03 shoot/explant) for the control treatment. As for shot length the longest shoot was obtained when kinetin was addied to the medium at concentration of 0.25 mg/L (6.66 cm in Camarosa cv.) followed by kinetin (6.61 cm in Rosalinda cv.).
The results from second and third experiments showed that survival percentage, shoots length, roots length, roots number, fresh and dry weight were greatly affected by adding NaCl to the MS medium. As the NaCl concentration was increased the proliferation rates were decreased. Moreover, plantlets of the three cultivars differed in there tolerance to the addition of NaCl.
Sweet Charlie plantlets and Camarosa plantlets seemed to be more tolerant than Rosalinda cultivar.
