Abstract

Summary and conclusions
In this thesis simple, reliable and sensitive spectrophotometric method was developed for the determination of eight cephalosporins namely cefotaxime, cephapirin, cephradine, cephalexin, ceftazidime, cefazoline, ceftriaxone and cefuroxime. In addition spectrofluorimetric method was also developed for determination of seven cephalosporins namely cefotaxime, cephapirin, cephradine, cephalexin, cefazoline, ceftriaxone and cefuroxime.
The thesis falls into five main parts
The first part
Includes general introduction about the medicinal importance of cephalosporins, information about chemical structure, mechanism of action and therapeutic indication. Also this part comprises a literature review of the reported analytical methods for determination of the studied drugs. These methods were classified into titrimetric, spectrophotometric, spectrofluorimetric, chromatographic, electrophoretic and electrochemical method. In this part also the scope of the work was explained.
The second part Includes kinetic study of the reaction of cefotaxime, cephapirin, cephradine, cephalexin, ceftazidime, cefazoline, ceftriaxone and cefuroxime with alkaline potassium permanganate followed spectrophotometric measurement. This part comprises two sections.
The experimental section includes the laboratory experimental information about all the instruments, drugs, reagents, solutions, pharmaceutical formulations and procedure used in this method.
The second section (Results and Discussion) include the results obtained from the analysis of the studied cephalosporins by the proposed method. In addition full details about the suggested reaction mechanism, kinetic of the reaction (initial rate and fixed time method), validation of the proposed method and application of this method for determination of studied drugs in pure form and pharmaceutical formulations were also described.
The developed kinetic method was applied successfully for determination of the studied drugs in their pharmaceutical dosage forms, the results obtained were validated by comparison with the reported methods by mean of F- and t- tests at 95 % confidence limit, and no significant difference were found indicating good accuracy and precision of the proposed method.
The third part Include kinetic spectrofluorimetric analysis of cefotaxime, cephapirin, cephradine, cephalexin, cefazoline, ceftriaxone and cefuroxime after their alkaline hydrolysis with 1.0 M sodium hydroxide. This part comprises into two sections

Also the proposed kinetic method was applied successfully for determination of cephalexin, cephradine, cefuroxime, ceftriaxone, cephapirin and cefazoline in real human plasma.
