Abstract

The present study aimed to examine if Punica granatum (pomegranate) natural extracts (PPME and PJ) could achieve protection against chlorpyrifos induced cardiotoxicity in male rats. This study was carried out using 36 male albino rats divided into 6 equal groups.All rats were fasted over night at the end of the study period and the animals were weighed and sacrificed under ether anesthesia at 8.00 in the morning. from each rat, two blood samples were collected. The first was taken on EDTA as anticoagulant for determination of hematological parameters and the second was centrifuged at 855xg for 10 minutes and sera were preserved at -200C for later biochemical analysis. Animals were dissected immediately after collecting blood and the hearts were removed, cleaned with saline solution (0.9%) and weighed. Relative weight of the each heart was calculated as the ratio of organ weight to animal body weight (mg/100 gb.wt.). Hearts were then divided longitudinally into two parts, one was homogenized in distilled water(1/10 weight/volume), centrifuged for 10 min. at 855×g. and the supernatants were transferred into eppendorf tubes and preserved at -80 ºC until used for various biochemical assays. The second was frozen at -80 Cº until used for Flow cytometry analysis. Heart samples from left parts were fixed in 10% neutral formalin for histopathological examination.In conclusion, the marked hypotention, hypolipidemic and antioxidant effects shown by pomegranate extracts greatly encourage the recommendation of its use as a therapeutic agent in people with cardiotoxicity, as well as assistant agent to enhance the effect of anti-toxicity drugs. However, further investigations about the effectiveness of many compounds isolated from this extract must be tested for cure or protection from cardiotoxicity and its accompanied complications.
