Abstract

This study was carried out to investigate the effect of cypermethrin on the metabolic profile in common carp (cyprinus carpio) using gas chromatography- mass chromatography (GC-MS) metabolomics approach. For this purpose, thirty six healthy common carp with average weight 25-30 g were distributed into 3 groups; six for each. The first group used as control, the second group exposed to 0.1 µg/L cypermethrin, the third group exposed to 1 µg/L cypermethrin for 24 and 96 hours. Samples of brain and hepato-pancrease tissues were taken after 24 and 96 hours and subjected to the metabolomics analysis using GC-MS to identify the metabolites peaks. The effect of cypermethrin on the brain metabolome resulted in significant rise in the level of tryptophan, histidine, glutamine, glutamate, aspartic acid, tyrosine, GABA and glycine levels in the exposed group related to control. Also, cypermethrin affected the TCA cycle via inhibition of its intermediates. Significant decreases were found in glucose, succinate, fumerate, malate levels in the exposed groups in compared to control. Regarding the essential amino acids, significant decrease in lysine and valine levels in the exposed groups in compared to control. Additionally, significant rise in the levels of alanine and serine with significant decline in the level of ornithine was observed in the exposed groups relative to control. Significant changes were observed after cypermethrin exposure on the purine metabolism. After exposure to cypermethrin for 24 hours, significant decline was detected in the level of inosine with significant disturbances in other metabolites related to purine metabolism. The results of hepatic metabolome revealed significant declines in the levels of all detected essential amino acids such as lysine, leucine, valine, histidine, tryptophan and methionine in exposed groups related to control. Significant reductions were found in the level of alanine, ornithine, oxoproline, glycine, serine, hydroxyproline and tyrosine levels with significant rise in asparagine and proline levels after cypermethrin exposure for 24 and 96 hours. After exposure to cypermethrin, significant decrease in the level of malate, lactate, pyruvate, fumerate and Succinate in cypermethrin exposed groups in comparison with control. The metabolites related to the purine metabolism were also altered by cypermethrin exposure. Significant reduction in xanthine, guanine and hypoxanthine were detected in the exposure groups compared to the control one after exposure to cypermethrin for 24 and 96 hours.
