Abstract

The present work was carried out to evaluate the pharmacological effect of Nigella sativa oil (400mg/kg) and Aloe vera gel aqueous extract (15ml/L) of drinking water in rabbits. Measurements were made at different periods (one day, one week and two weeks) after administration to follow up the induced effects of the medicinal plants on some haematological, biochemical parameters and their antioxidant activity on rabbits. 1- The medicinal plants: a- Nigella sativa oil. b- Aloe vera gel aqueous extract. 2- Experimental animals : This study was performed on 15 rabbits of either sex (1.5±.5Kg B.wt.)The animals were housed under standard laboratory conditions in polypropylene cages, provided with food and water ad libitum. The animals were acclimatized to the laboratory environment for at least one week before the experimental session. The experimental protocol was approved by Institutional Animal Ethical Committee. Rabbits were divided in to 3 experimenal groups (each of 5 rabbits). 1- The first group as a control group (G1): fed on a commercial diet without any additives. 2- The second group (G2): fed on a commercial diet+ 400 mg/Kg Nigella sativa oil given orally. 3- The third group (G3): fed on commercial diet+ Aqueous extract 10%w ⁄v of 15ml ⁄ L Aloe vera gel. 3-Sampling and experiments : The blood samples were collected after one day, one week and two weeks of treatment. Blood was collected from the ear vein on EDTA for hematological studies as red blood cell count(RBCs), White blood cells count(WBCs), Differential leukocytic count ,Platelet count, Hb concentration and PCV% were determined. Another sample of blood were collected and allowed to clot for serum separation by centrifugation at 3000rpm for 10min for biochemical studies as liver function test (ALT and AST), Kidney function test (serum urea and creatinine) and serum antioxidant enzymes(reduced glutathione, malondialdehyde and catalase). Data were analyzed using the statistical package for social science (SPSS), 15.0 software, 2008). 4- Effects on blood picture : The results of the experiment showed that There were insignificant changes in RBCs, Hb, PCV, WBCs and platelets. 5- Biochemical determination : a-Effects on liver function parameters serum (ALT and AST) : The results of ALT and AST showed non- significant changes in the treated groups when compared with the control group. b-Effects on kidney function parameters(serum urea and creatinine) : The results of urea and creatinine showed non- significant changes in the treated groups when compared with the control group. c-Evaluation of serum oxidative stress and antioxidant enzymes : - Effects on serum catalase : Nigella sativa and aloe vera gel groups evoked a significant increase in catalase and aloe vera gel after one day and one week of treatment compared to control group, While the nigella sativa and aloe vera gel groups elicited non-significant difference after two weeks of therapy compared to control group. - Effect on serum reduced glutathione (GSH) : The present work mirrored a significant increase in reduced glutathione after one day and one week in the group was treated with aloe vera gel and non-significant change in aloe vera gel and control groups. Also there was a significant increase in reduced glutathione in nigella sativa and aloe vera gel treated groups in comparison with control group after two weeks. - Effects on serum malondialdehyde (MDA) : MDA results revealed a significant decrease in nigella sativa and aloe vera gel groups during the first day of treatment when compared with control group, While the nigella sativa and aloe vera gel groups elicited non-significant difference after one week and two weeks of therapy compared to control group.
