Abstract

the effect of non-steroidal anti-inflammatory drugs is a matter of debate. Many researchers found significant associations between both. Aim of this study : This study was conducted to evaluate the effects of non-steroidal anti-inflammatory drugs with different concentrations on proliferation and osteogenic capability of dental pulp stem cells. Materials and Methods : Dental pulp stem cells (DPSCs) harvested from pulp tissue of extracted lower left third molar treated with diclofenac sodium (10-6 and 10-4 M), acetylsalicylic acid (10,50 and 200 μg/mL) and meloxicam (0.01,0.1 and 1 μM) in 96-well plates for 1,3 and 5 days. Drug effects on proliferation and cell viability were tested using the MTS assay. The osteogenic capability of dental pulp stem cells was tested using an osteogenic medium for 1 weeks using concentrations of 10 μg/mL acetylsalicylic acid, 10-6 M diclofenac sodium and 0.1 μM meloxicam. Results : It was found that low concentrations of ASA 10-50 μg/mL enhance the proliferation and growth of dental pulp stem cells and also improve the osteogenic capability. Also, diclofenac sodium at 10-6 M at day 1 had higher proliferative effect of on DPSCs. Moreover, diclofenac sodium at concentration 10-6 M promotes the osteogenic ability of DPSCs. 0.1and 1 μM meloxicam showed a more cell proliferation on day 3 than other days also osteogenic capability of 0.1 μM meloxicam with osteogenic medium. Conclusion : based on our results NSAIDs, tested in therapeutically used concentrations, don’t interfere with the DPSCs capability of proliferation and cell growth. Also, NSAIDs don’t exert any inhibitory effect on osteogenic differentiation.
