Abstract

The purpose of this study was to investigate how addition of L arginine to either maturation or vitrification medium affected the developmental competence of vitrified/thawed immature buffalo oocytes. In this study, different concentrations of L arginine (50, 100, and 250 ul/ml) were used. The results concluded that the addition of Larginine at concentration of 250 µL/ml to maturation medium enhance cleavage and morula formation. At the same time the vitrified/thawed oocytes that were vitrified in vitrification medium supplemented with L-Arginine in a concentration of 250 µL/ showed the highest morula formation rate mmol The vitrified/thawed oocytes that were vitrified and matured in medium supplemented with L-Arginine in a concentration of 50 µL/ml and 100 µL/ml showed higher cleavage and morula development rate than oocytes vitrified and matured in medium supplemented with L-arginine in concentration of 250 µL/ml . Also the control group that was vitrified and showed in medium without supplementation with L arginine showed less cleavage and morula formation rates. We concluded that supplementation of either verification or maturation medium with L arginine in a concentration of 250 µl/ml improve the cleavage and morula formation rate in comparison with a concentration of 0, 10 and 100 µl/ml. but supplementation of both vitrification and maturation medium with L arginine at a concentration of 50 and 100 µl/ml improved the cleavage and morual development rate in comparison with concentration of 0 or 250 µl/ml.
