Abstract

This work mainly aims at the efficiency of some medicinal plants in controlling and treatment of some pathogenic bacteria in a trial to decrease the used medicines with high harmful side effects. Obtaining some human pathogenic bacteria from MU hospital. Purification and identification of the obtained pathogenic bacteria. Morphological, biochemical and molecular identification of screened pathogenic bacteria.C ollection of some medicinal plants (Farsetia aegyptia Turra., Fagonia mollis Delile., Lactuca serriola L and Symphotrichum squamatum (Spreng.) Nesom.) grown naturally in Egyptian habitats. P reparation of plant extracts and phytochemical analysis of selected plant species Purification and characterization of the most active compounds by GC-MS. Screening the prepared extracts and active constituents for their biological activities : antimicrobial activity, antioxidant, and antitumor potentiality. A total of 100 clinical samples were collected during the 2021 year from Mansoura University Hospital. The clinical samples included blood (30), urine (40), and stool (30) from different patients with different genders and ages ranging from 16 to 65 years. The studied isolates were differences with other in colony morphology. A large variety of both Gram-negative and Gram-positive bacteria are involved. This bacterial species is a member of the ESKAPE group recently described as the main contributor to the health human infection problem. The results of the Vitek®2 system showed that 100 isolates were identified.The qualitative analysis showed that plant samples were rich in varying amounts of alkaloids, flavonoids, phenols, saponins, and tannins and trace quantities of glycosides and steroids, anthraquinones, terpenoids. The chemical constitutes of the extracted Lactuca serriola, Fagonia mollis, and Symphotrichum squamatum were established by Gas-Chromatography Mass Spectroscopy “GC-MS”. In Fagonia mollis analysis confirmed that 19 components were characterized, and 2-Ethylhexanoic acid is situated as the major component with 34.23% composition. In Lactuca serriola, 10 components were characterized, and Lup-20(29)-en-3β-ol, acetate (42.62%) and Germanicol (36.01%) are situated as the major componentf. On the other hand, in Symphotrichum squamatum, 13 components were characterized 2-Ethyl-hexanoic acid is situated as the major component with 19.32% composition. In this work, the antioxidant properties of four selected plant extracts were in vitro evaluated by DPPH assay. In the present study, the antibacterial activity of the selected plant species from methanol was assessed in vitro antimicrobial susceptibility testing. The inhibitory zone of methanolic extracts of selected plant differed depending on the type of bacteria studied. The results showed that the extracted shoot of these plants have moderate cytotoxic activity for S. squamatum (HepG-2 and PC3) and L. serriola (HepG-2), whereas F. aegyptia and F. mollis have weak cytotoxic activity for two selected human tumor cells. In addition, all the tested samples revealed non-cytotoxic activities against the normal lung fibroblast (WI-38) cell line, with IC50 >100 µg/mL.
